浙江湖州二化螟种群对七种杀虫剂的抗性监及常用杀虫剂的田间药效评估
摘要: 【目的】本研究旨在探究浙江省湖州市二化螟Chilo suppressalis种群对常用杀虫剂的抗性特征及抗性位点突变频率,结合田间试验系统评估环丙氟虫胺等新型药剂的防控潜力,为太湖流域单季晚稻区制定二化螟抗性治理策略提供科学依据。【方法】通过稻苗浸渍法和毛细管点滴法测定浙江湖州二化螟种群对4类7种杀虫剂[双酰胺类(氯虫苯甲酰胺、环丙氟虫胺和溴虫氟苯双酰胺)、 双酰肼类(甲氧虫酰肼)、 大环内酯类(乙基多杀菌素和阿维菌素)和有机磷类(三唑磷)]的敏感性; 采用基因测序技术检测抗性个体鱼尼丁受体基因突变位点; 通过在单季晚稻破口期对40%氯虫·噻虫嗪水分散粒剂(water dispersible granule, WG)、 20%环丙氟虫胺悬浮剂(suspension concentrate, SC)、 200 g/L四唑虫酰胺SC+5%阿维菌素乳油(emulsifiable concentrate, EC)、 10%阿维·甲虫肼微囊悬浮剂(capsule suspension, CS)和25%乙基多杀菌素WG 7种药剂实施人工喷药的随机区组的小区试验,来评估其对二化螟的田间药效。【结果】抗药性监测显示:二化螟湖州种群对氯虫苯甲酰胺产生高水平抗性(抗性倍数278.0),对乙基多杀菌素、甲氧虫酰肼、阿维菌素和三唑磷呈中等抗性,抗性倍数分别为79.2, 51.7, 24.3和10.3。从鱼尼丁受体基因中鉴定出多个与氯虫苯甲酰胺抗性相关的突变。鱼尼丁受体基因检测发现与氯虫苯甲酰胺抗性相关的突变位点Y4667C突变频率为21.1%(均为杂合型),而I4758M突变频率高达84.2%。田间试验结果表明,单季晚稻破口期施药1次后, 20%环丙氟虫胺SC保苗效果(94.66%)和幼虫防治效果(98.57%)均显著优于其他药剂(25%乙基多杀菌素WG保苗效果80.85%、幼虫防治效果85.93%, 200 g/L四唑虫酰胺+5%阿维菌素EC保苗效果66.43%、幼虫防治效果71.87%, 10%阿维·甲虫肼CS保苗效果63.84%、幼虫防治效果58.83%)。40%氯虫·噻虫嗪WG在水稻扬花期施药时的二化螟残留虫量较水稻破口期施药时降低了58.3%。【结论】湖州二化螟种群已对氯虫苯甲酰胺产生严重抗性,建议浙北稻区暂停使用该药剂,对中等抗性药剂实施轮换使用策略。环丙氟虫胺可作为优势替代药剂,推荐在太湖流域第3代二化螟防治中采用“破口期+扬花期后”的二次施药方案以提升防治效果的持续性。

Abstract: 【Aim】 This study aims to explore the resistance characteristics and mutation frequency of resistance sites of Chilo suppressalis populations in Huzhou City, Zhejiang Province, eastern China to common insecticides, and to systematically evaluate the control potential of new pesticides such as cyproflanilide through field trials, thereby providing a scientific basis for formulating resistance management strategies in single-cropping late rice regions of the Taihu Lake Basin. 【Methods】 The susceptibility of C. suppressalis populations to seven insecticides of four categories including diamides (chlorantraniliprole, cyproflanilide and broflanilide), diacylhydrazines (methoxyfenozide), milbemycins (spinetoram and abamectin), and organophosphates(triazophos)was determined using rice seedling dipping and capillary tube microapplication methods. The mutation sites in the ryanodine receptor gene of resistant individuals were analyzed through gene sequencing. A randomized plot trial was conducted to evaluate the field efficacy of seven insecticides, including 40% chlorantraniliprole·thiamethoxam water dispersible granule (WG), 20% cyproflanilide suspension concentrate (SC), 200 g/L tetraniliprole SC+5% abamectin emulsifiable concentrate (EC), 10% abamectin· methoxyfenozide capsule suspension (CS), and 25% spinetoram WG, which were artificially sprayed during the booting stage of single-cropping late rice, against C. suppressalis.【Results】 Resistance monitoring revealed that the Huzhou population of C. suppressalis exhibited high-level resistance to chlorantraniliprole (resistance ratio: 278.0) and moderate resistance to spinetoram, methoxyfenozide, abamectin and triazophos, with the resistance ratios of 79.2, 51.7, 24.3 and 10.3, respectively. Multiple chlorantraniliprole resistance-associated mutations were identified in the ryanodine receptor gene. Testing of the ryanodine receptor gene revealed the mutation frequency of 21.1% at the Y4667C locus (all heterozygous) associated with chlorantraniliprole resistance, while the I4758M mutation frequency was as high as 84.2%. Field trials demonstrated that 20% cyproflanilide SC exhibited significantly superior plant protected efficacy (94.66%) and control efficacy against larvae (98.57%) to other agents (plant protected efficacy and control efficacy against larvae: 80.85% and 85.93% for 25% spinetoram WG, 66.43% and 71.87% for 200 g/L tetraniliprole SC+5% abamectin EC, and 63.84% and 58.83% for 10% abamectin·methoxyfenozide CS, respectively) when applied once at the booting stage of single-cropping late rice. Application of 40% chlorantraniliprole·thiamethoxam WG during the rice flowering stage reduced residual C. suppressalis populations by 58.3% relative to its application at the rice booting stage. 【Conclusion】 Huzhou C. suppressalis populations has developed severe resistance to chlorantraniliprole. Immediate discontinuation of this insecticide in northern Zhejiang rice areas is recommended, with rotational use strategies for moderately resistant agents. Cyproflanilide shows potential as a superior alternative, and a dual-application strategy (“booting stage+post-flowering stage”) is proposed to enhance control efficacy sustainability against the 3rd-generation C. suppressalis in the Taihu Lake Basin.
